A Cell Permeable NPE Caged ADP-Ribose for Studying TRPM2
Transient potential receptor melastatin-2 (TRPM2) is a non-selective Ca(2+)-permeable cation channel of the TRPM channel subfamily and is mainly activated by intracellular adenosine diphosphate ribose (ADPR). Here we synthesized a 1-(2-nitrophenyl)ethyl caged ADPR (NPE-ADPR) and found that uncaging of NPE-ADPR efficiently stimulated Ca(2+), Mg(2+), and Zn(2+) influx in a concentration-dependent...

Fast solute release from photosensitive liposomes: an alternative to 'caged' reagents for use in biological systems.
The kinetics of release of soluble marker trapped in liposomes of gel phase phospholipid containing a photoisomerisable phospholipid analogue have been investigated. Marker release is triggered by UV laser flash photolysis at 355 nm. A markedly temperature-dependent release rate is seen, and above 25 degrees C millisecond release kinetics can be achieved. These results suggest that such liposom...
متن کاملCaged circular antisense oligonucleotides for photomodulation of RNA digestion and gene expression in cells
We synthesized three 20 mer caged circular antisense oligodeoxynucleotides (R20, R20B2 and R20B4) with a photocleavable linker and an amide bond linker between two 10 mer oligodeoxynucleotides. With these caged circular antisense oligodeoxynucleotides, RNA-binding affinity and its digestion by ribonuclease H were readily photomodulated. RNA cleavage rates were upregulated ~43-, 25- and 15-fold ...
متن کاملBiological containment facility for studying infectious disease.
To effectively characterize newly recognized viruses (Marburg, Lassa, etc.) and to study other highly virulent infections for which no effective prophylaxis or therapy exists, special containment facilities must be utilized and conventional techniques modified to minimize risk to laboratory personnel. This paper describes a laboratory suite for such studies, contained within a larger research f...
